The level of interleukin-9 (IL-9) in the specimens was determined by double antibody sandwich assay. The microporous plate was coated with purified rat interleukin 9 antibody to prepare a solid phase antibody, and rat interleukin 9 was sequentially added to the microcapsule of the coated monoclonal antibody, and then bound with HRP-labeled IL-9 antibody. The antibody-antigen-enzyme-labeled antibody complex was formed, and after thorough washing, the substrate TMB was added for color development. TMB is converted to blue under the catalysis of HRP enzyme and converted to the final yellow color by the action of an acid. The color depth is positively correlated with IL-9 in the sample. The absorbance (OD value) was measured at 450 nm using a microplate reader, and the concentration of rat interleukin 9 (IL-9) in the sample was calculated from a standard curve.
