Human osteopontin (OPN) enzyme-linked immunosorbent assay (ELISA) kit instruction manual

The kit employs a double-antibody sandwich immunoassay to quantify the concentration of human osteopontin (OPN) in a given sample. The process begins with a microtiter plate pre-coated with a purified monoclonal antibody specific to OPN, forming a solid-phase capture antibody. After incubation, the sample is added, allowing OPN to bind to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then introduced, which binds to the captured OPN, forming a stable antibody-antigen-enzyme complex. Following a thorough wash to remove unbound components, a substrate solution containing TMB (3,3',5,5'-tetramethylbenzidine) is added. Under the catalytic action of HRP, TMB is oxidized to a blue product, which is then stopped by adding an acidic solution, resulting in a yellow color. The intensity of the yellow color is directly proportional to the amount of OPN present in the sample. The optical density (OD) is measured at 450 nm using a microplate reader, and the OPN concentration is determined by comparing the OD value to a standard curve generated from known concentrations of OPN. This method ensures high specificity, sensitivity, and reproducibility, making it ideal for clinical and research applications.
